Discovery of the mechanism of traditional Chinese medicine monomer notopterygiol inhibiting JAK2 / 3 and treating rheumatoid arthritis

 

On September 15, 2020, researcher Hongyan Wang of the Chinese Academy of Sciences, Professor Yongjun Wang and researcher Qianqian liang of Longhua Hospital Affiliated to Shanghai University of traditional Chinese Medicine published an article in the journal Cell reports: The natural compound Notopterol binds and targets JAK2/3 to ameliorate inflammation and arthritis,It was found that notopterygiol, a monomer of traditional Chinese medicine, was an inhibitor of JAK2 / 3 and its mechanism of action in the treatment of rheumatoid arthritis (RA)

Rheumatoid arthritis (RA) is a common autoimmune disease in clinic. Patients with rheumatoid arthritis mainly present with joint pain, swelling and irreversible joint deformities.

 

 
 

The study found through clinical randomized controlled trials thatThe classic prescription "Juanbi Decoction" has curative effect on rheumatoid arthritisAnd has the effect of treating collagen induced rheumatoid arthritis (CIA) mouse model and tumor necrosis factor transgenic mouse (TNF TG) model. Later, researchers detected that "Juanbi Decoction" contained notopterygiol by liquid chromatography-mass spectrometry.

 

 

In this study, researchers constructed CIA mouse models with dba/1j background and c57/bl6 background, and confirmed that notopterygiol has the effect of treating RA by oral and intraperitoneal administration, respectively. . RNA SEQ, small molecule protein interaction technology and a series of screening,.And it was confirmed in zebrafish system that notopterygiol exerts the function of inhibiting inflammation by regulating jak2/3. Using the mouse RA model, it was confirmed that the effect of combined use of notopterygiol and enli (TNF inhibitor), a commonly used drug in clinic, was better. In conclusion,Notopterygiol exerts its therapeutic effect on rheumatoid arthritis by targeting JAK2 / 3-statsignaling pathwayThis also puts forward a new strategy for the follow-up study on whether notopterygiol can treat other diseases with JAK2 / 3-statsignaling pathway abnormalities.

New findings of aucubin in alleviating experimental brain injury

 

Recently, Professor hangchunhua and Dr. Li Wei from the Department of Neurosurgery, Nanjing Drum Tower Hospital, Affiliated Hospital of Nanjing University School of medicine, conducted a study on aucubin attenuating oxidative stress and inflammatory response of experimental brain injury through Nrf2 mediated signaling activity. The article was titled "aucubin alleles oxidative stress and inflammation via Nrf2 mediated signaling activity in experimental traumatic brain injury", and was published in the Journal of neuroinflammation (if=5.793, https://doi.org/10.1186/s12974-020-01863-9 )Upper.

 

 

Aucubin (AU) is a natural plant iridoid glycoside with antioxidant and anti-inflammatory activities, but its role in traumatic brain injury (TBI) model is still unclear.This study explored the potential role of Au in H2O2 induced oxidative damage of primary cortical neurons and weight drop induced TBI Model in mice.In vitro experiments, different concentrations of Au (50 μ g/ml, 100 μ g/ml or 200 μ g/ml) were added to neurons 0h and 6h after H2O2 (100 μ m) stimulation. After 12h, neuronal cells were collected for immunoblotting (WB), immunofluorescence, and m29,79-dichlorodihydrofluorescein diacetate (DCFH-DA) staining. In vivo experiments, Au (20mg/kg or 40mg/kg) was injected intraperitoneally at 30min, 12h, 24h and 48h after modeling. Brain water content, neurological deficit and cognitive function were measured at specific time. . RNA interference experiment was used to study the effect of nuclear factor erythroid 2-related factor 2 (Nrf2) on Au (40mg/kg) in TBI mice treated with Au (40mg/kg). The lentivirus was injected into the lateral ventricle of mice 72h before TBI was established. The cortex was removed 72h after TBI for WB and Q-PCR.

 

. In the TBI mouse model, Au significantly alleviated brain edema, histological injury, and improved neurological and cognitive dysfunction. Au significantly inhibited HMGB1 mediated sterile inflammation. Nrf2 knockout in TBI mice attenuated the antioxidant and anti-inflammatory neuroprotective effects of Au. To sum up,Studies have shown that AU has neuroprotective effects in a mouse model of TBI by inhibiting oxidative stress and inflammatory response; the mechanism involves triggering the Nrf2 induced antioxidant system.

Progress in the absorption characteristics of nonvolatile components in Amomum villosum water extract

 

Amomum villosum, a traditional Chinese medicine, is the dried and mature fruit of Amomum villosum, green shell sand or Hainan sand in the ginger family. It has the effects of dissolving dampness and moving Qi, warming and stopping diarrhea, and relieving pregnancy. It mainly treats qi stagnation in the spleen and stomach, dampness obstructing the middle energizer, deficiency and cold in the spleen and stomach, vomiting and diarrhea, restless fetal movement, malignant obstruction of pregnancy and other effects. However, it is unclear whether the nonvolatile active ingredients in Amomum villosum water extract are helpful in the treatment of gastrointestinal diseases. Recently, zouhuiqin and yanyonghong research team of Beijing University of traditional Chinese Medicine published a paper titled "the absorption characteristics of nonvolatile components in a water extraction from Amomi Fructus as determined by in situ single pass internal perfusion and high performance liquid chromatography" in "Frontiers in Pharmacology"Absorption characteristics of nonvolatile components in aqueous extract of Amomum villosumProgress has been made in research.

 

 

. The team identified 19 components in Amomum villosum water extraction, which showed different absorption characteristics in different intestinal segments, among which five compounds had negative Peff values in the jejunum. Subsequently, the team used HPLC to quantitatively study the six components of catechin, vanillic acid, epicatechin, polydatin, Isoquercitrin and quercitrin.The study suggested that the effective permeability and intestinal absorption site of nonvolatile components could be better evaluated using the rat orthotopic unidirectional intestinal perfusion model.The results laid a foundation for elucidating the pharmacodynamics of Amomum villosum.
