CORYDALIS BUNGEANAE HERBA
This product is the dried whole grass of Corydalis bungeana turcz., a plant of Papaver family. Harvest in summer, remove impurities and dry in the sun.
[properties] the product is crumpled into a mass, 10-30cm long. The taproot is conical, and the surface is brownish yellow. The stem is thin, multi branched, gray green or yellowish green on the surface, with 5 longitudinal edges, soft, hollow section. The leaves are mostly shrunken and broken, dark green or grayish green, and the intact leaves are two to three times pinnately cleft. Flowers are rare, corolla lip-shaped, spurred, lilac. The capsule is oblate, oblong, and podlike. The seeds are oblate, black and glossy. The breath is slight and the taste is bitter.
. The cortical parenchyma cells were irregular in shape, with 7-10 rows of thick horn cells at the ridge. The central column sheath is 1-2 rows of fibers, arranged in a ring, and the fibers at the ridge are arranged in a half moon shape. The outer tough vascular bundle is located at the ridge, the phloem is narrow, and the xylem is composed of vessels, tracheids, fibers and parenchyma cells. The medulla is broad with a large cavity in the center.
(2) Take 0.5g of this product powder, add concentrated ammonia test solution to moisten it, add 10ml of trichloromethane, place it overnight, filter it, evaporate the filtrate to dryness, add 11ml of trichloromethane to dissolve the residue as the test solution. In addition, take the reference substance of corydalis and add methanol to make a solution containing 1mg per 1ml as the reference solution. According to the test of thin-layer chromatography (general rule 0502), suck 10 μ l of each of the above two solutions, dot them on the same silica gel G thin-layer plate prepared with 0.4% sodium hydroxide solution, use cyclohexane trichloromethane methanol (7:2:1) as the developing agent, develop, take out, dry, and spray with dilute bismuth potassium iodide test solution. In the chromatogram of the test sample, spots with the same color appear at the corresponding position of the chromatogram of the control sample.
[inspection] impurities shall not exceed 2% (general rule 2301).
.
[extract] according to the hot leaching method under the determination method of water-soluble extract (general rule 2201), it shall not be less than 18.0%.
[content determination] determine according to high performance liquid chromatography (general rule 0512).
Chromatographic conditions and system suitability test octadecylsilane bonded silica gel was used as filler; The mobile phase was methanol-0.015mol/l phosphate buffer (ph6.7) (take 1.58g of potassium dihydrogen phosphate and 0.76g of potassium dihydrogen phosphate, add 1000ml of water to dissolve, mix well, and adjust the pH value to 6.7 with sodium hydroxide test solution) (70:30); The detection wavelength was 289nm. The number of theoretical plates should not be less than 6000 according to the violin peak.
Preparation of reference solution take a proper amount of Corydalis reference, weigh it accurately, add methanol to make it with 40% sodium hydroxide per 1ml? G solution.
Preparation of the test solution take about 0.5g of the powder (passing through No. 3 screen), accurately weigh it, place it in a corked conical flask, precisely add 25ml of methanol, close the stopper, weigh it, soak it for 1 hour, sonicate it (power 250W, frequency 33khz) for 30 minutes,
Take it out, cool it, weigh it again, make up the lost weight with methanol, shake it well, filter it, and take the filtrate.
The determination method is to precisely suck 20 μ l of the reference solution and 20 μ l of the test solution, inject them into the liquid chromatograph, and determine.
The content of Corydalis (c21h21o5n) in this product shall not be less than 0.14% calculated as a dry product.
