Rabdosia rubescens is a dry whole herb of parvium, a plant belonging to the genus rabdosia. It has the effects of anti-inflammatory and detoxification, anti-inflammatory and analgesic, anti-tumor, etc. it is used to treat sore throat and tonsillitis, and can relieve symptoms and prolong survival time for patients with esophageal cancer, liver cancer, lung cancer and other cancers. Oridonin is a diterpenoid compound extracted from Rabdosia rubescens, which accounts for more than 90% of the effective components of Rabdosia rubescens. Oridonin is the main anti-tumor active component. Oridonin has obvious inhibitory effect on the growth of many human cancer cell lines. So why can oridonin play an anti-tumor role? It is found that oridonin is mainly realized through the following five points.

    First, inducing cancer apoptosis.

Second, block the proliferation cycle of tumor cells. The disorder of cell cycle regulation mechanism is an important reason for uncontrolled cell proliferation and carcinogenesis. Oridonin can significantly inhibit the growth of tumor tissue by blocking the proliferation cycle of cancer cells.

Third, reduce telomerase activity. Telomerase is a special ribonucleoprotein polymerase, which can use its RNA as a template to synthesize telomeres, so as to make up for the shortening of telomeres caused by the replication process, prolong the life of cells and even immortalize. There is a high correlation between telomerase and tumor. It was found that oridonin could down regulate the telomerase activity of related tumor cells, thus inhibiting cancer cells.

Fourth, antimutagenic effect. The anti mutation mechanism of oridonin may be caused by the following factors: extracellular effect, oridonin blocks or inactivates the mutagenic activity of the mutagen outside the cell; intracellular effect, oridonin blocks the effect of the mutagen on the target molecule through a certain mechanism. Oridonin itself has no biological effect on normal cells.

Fifth, enhance the efficacy of other anti-tumor drugs. Oridonin has attracted much attention because of its obvious cytotoxicity and small side effects. Oridonin is limited in application due to its poor water solubility. Therefore, oridonin has been modified to improve its water solubility and activity, and developed into a new anti-tumor drug to make it an important research direction.

Oridonin has a significant inhibitory or killing effect on leukemia, breast cancer, melanoma and other tumors. It is mainly used in the treatment of anti-tumor, antibacterial, heat clearing and detoxification, anti-inflammatory and analgesic, stomach invigorating and blood activating. However, because it is slightly soluble in water and tastes extremely bitter, its clinical application is limited to a great extent. As a new drug delivery system, in recent years, scholars have carried out the research and exploration of oridonin nano preparations, including oridonin β - cyclodextrin inclusion complex, nanoparticles, liposomes, microemulsion polymer micelles and nano suspensions. It is found that nano preparation can effectively improve the water solubility of oridonin, improve its bioavailability, andOridoninIt has development value and broad application prospects.
